Traditional PGD methods require family-specific and locus-specific designs that are time consuming (several months) and labour intensive. In addition, using these methods, segregation of only one risk allele at a time can be traced. In addition, direct mutation detection assays may suffer from artefacts, such as allele drop out, and consequent misdiagnosis. Moreover, these methods may produce carrier offspring, e.g. in the case of X-linked disorders or balanced translocation carriers. Our patented strategy is a linkage-based approach that enables a generic concurrent PGD and PGS and overcomes the above-mentioned pitfalls of the traditional PGD methods. Using haplarithmisis we do not type the mutation per se, but rather scrutinize the flanking markers and impute the risk alleles. EXIR's PGD solution makes use of high-throughput genotyping data, e.g. SNP-array or next-generation sequencing technologies, and simultaneously reconstruct haplotypes and determines copy number states of the loci across the genome. Thus, by applying a single assay we can perform PGD and PGS simultaneously. Importantly, we can trace the segregational or mechanistic origin of genomic anomalies. This allows us to detect if a certain aberration occurs in meiosis or mitosis.
